What is 5-Amino-1-MQ?
5-Amino-1-methylquinolinium is a synthetic small molecule studied as an inhibitor of the enzyme nicotinamide N-methyltransferase (NNMT). Researchers use it to investigate how changing enzyme activity affects metabolism. Cell and mouse research.
What Does NNMT Do?
NNMT transfers a small chemical unit called a methyl group from SAM (S-adenosylmethionine) to nicotinamide. This forms 1-methylnicotinamide (1-MNA) and SAH (S-adenosylhomocysteine).
That reaction links nicotinamide metabolism with the cell’s supply of methyl donors. NNMT is a normal enzyme found in several tissues; its effects depend on the biological setting. NNMT chemistry and background.
How Inhibition Could Change the Pathway
- Reducing NNMT activity can reduce conversion of nicotinamide into 1-MNA.
- This may leave more nicotinamide available for recycling into NAD⁺ through the salvage pathway.
- It can also change SAM availability, connecting NNMT inhibition with methyl-group metabolism.
NAD⁺ participates in energy metabolism and other cellular reactions. Its concentration alone does not tell us how much ATP a cell produces or how energetic a person feels.

What Did Researchers Measure?
Neelakantan and colleagues’ study appeared online in November 2017 and in the January 2018 issue of Biochemical Pharmacology.
Cultured Fat Cells
The researchers reported lower 1-MNA, higher NAD⁺ and SAM, and reduced lipid accumulation with NNMT inhibition. These were measured cellular findings, rather than purely hypothetical changes.
Mice with Diet-Induced Obesity
In an 11-day experiment with nine mice per group, injected 5-Amino-1MQ reduced body weight, white fat mass and fat-cell size without a significant change in total food intake. Read the original study and figures.
These findings from cultured cells and a short mouse study point researchers toward NNMT inhibition as a metabolic pathway worth continued investigation — with animal-model and injection-route factors that any follow-on research (oral, longer-duration, human) would need to account for.
